中药姜黄对黑腹果蝇肿瘤迁移模型的影响

摘要:姜黄,作为临床上广泛使用的一味活血化瘀类药物,其在改善微循环、改变血液流变性等方面的作用研究较为明确。中医基础理论认为血瘀与肿瘤的发生发展密切相关,但姜黄对于肿瘤生成及转移有何作用及其潜在调控机理尚未明确,仍需进一步探究。本研究选取黑腹果蝇为模式生物,建立细胞迁移和肿瘤侵袭模型,在基础培养基中添加姜黄水提物,对上述模型进行用药干预,明确其在体内对肿瘤细胞转移的影响。研究结果表明,适宜浓度的姜黄提取液可以显著抑制果蝇三龄幼虫期翅成虫盘中的细胞迁移。此外,姜黄还可大幅降低果蝇成虫eyeful眼部肿瘤的侵袭比率。综合上述结果,本文充分证实了姜黄在抑制细胞迁移和抗肿瘤转移方面的积极作用,为癌症等相关疾病的临床治疗和寻找潜在的药物靶点提供了新的思路。

Effects of Jianghuang on cell migration and tumor invasion in Drosophila
Abstract: Jianghuang (Curcumaelongae Rhizoma), a Traditional Chinese Medicine (TCM) herb mainly for treating blood stasis syndrome, has been widely investigated and applied in clinic, specially in improving microcirculation and changing hemorheology indexes. Meanwhile, blood stasis is an essential factor for tumorigenesis and metastasis during in the TCM basic theory system. However, the e?ects of Jianghuang on tumor growth and invasion, and the underlying mechanisms are still largely unknown. Here, we employed well-established Drosophila cell migration and tumor invasion models to explore whether Jianghuang has the anti-cancer activity in vivo. Our work has demonstrated that Jianghuang could strongly imped the ptc>scrib-IR induced cell migration at the 3rd instar larval stage. In addition, Jianghuang also significantly reduces the adult fly eyeful ocular tumor invasion at the moderate concentrations. Together, these evidences reveal a positive role of Jianghuang in suppressing cell migration and tumor metastasis, providing the potential drug targets and key clues for cancer clinical treatment strategies.
