手动供能的离心微流控芯片检测马拉硫磷

摘要:在资源有限的地区,采用无需电力的便携化装置检测有机磷农药对于食品安全是非常重要的。受拉线陀螺的启发,我们发展了一种手动供能的离心微流控芯片用于有机磷农药马拉硫磷的快速便携化检测。将乙酰胆碱酯酶(AChE)和显色剂吲哚乙酸酯(IDA)经普鲁兰多糖包封成膜后,分别预存储在微流控芯片的酶抑制区和显色区。样品加入酶抑制区反应一段时间后,将芯片插入拉线转盘中,拉动转盘产生的离心力使酶抑制区中的溶液转移到显色区,最后观察显色区的颜色以判断样品中有机磷农药的含量。该装置检测马拉硫磷的线性范围为0.1-20 μmol/L,检测限为0.1 μmol/L。此装置采用手动供能,无需外部电源,有望在现场检测中得到应用。

Hand-powered centrifugal microfluidic chip fot detecting marathion
Abstract:Detecting organophosphorus pesticide by a portable device that do not require electricity is very important for food safety in areas with limited resources. Inspired by the whirligig, we developed a hand-powered centrifugal microfluidic chip for rapid and portable detection of the organophosphorus pesticide, malathion. We encapsulated acetylcholinesterase (AChE) and chromogenic agent indole acetate (IDA) within pullulan to form the films and pre-stored them in the enzyme inhibition zone and the color development zone of the microfluidic chip, respectively. After adding the sample to the enzyme inhibition zone for a period of time, we inserted chip into the disc. Under the centrifugal force generated by pulling the disc, the solution in the enzyme inhibition zone was transferred to the color development zone. Finally, we measured the content of organic phosphorus pesticides in the sample by observing the color intensity of the color development zone. The linear detection range of the device to detect marathion is 0.1-20 μmol/L, and the detection limit is 0.1 μmol/L. The hand-powered device without external power supply has important application potential in field detection.
